Can Lab-Grown Antibodies Cure COVID-19?

Doctors are seeking to grow some of the body’s most potent disease-fighting weapons in a test tube and deploy them against COVID-19.

The treatment, known as monoclonal antibody therapy, could serve as both a remedy for patients infected with the coronavirus that causes COVID-19 and a way to prevent infection in frontline health workers and other people at high risk.

At least six companies expect to be testing antibodies on COVID-19 patients this summer.

Monoclonal antibodies have proven to be powerful tools against ailments including cancer and rheumatoid arthritis. A new treatment against Ebola is under review at the U.S. Food and Drug Administration.

Antibodies have advantages over conventional drugs that put them near the front of the pack of new therapies under development against COVID-19. For one, producing them is relatively fast, thanks to technology developed in the last 10 to 15 years.

“The whole process, from getting a blood sample to having antibodies ready for the clinic, was about three months,” said antibody sciences director Laura Walker at biotech company Adimab.

While safety testing is just beginning, Walker said antibody treatment is generally fairly safe.

“Everybody makes antibodies naturally,” she noted.

And since they are based on a well-studied part of the body’s defenses, Walker added, “the odds that a monoclonal antibody will be effective is quite high.”

Antibodies are a key part of the immune system. They work by attaching themselves to an invading germ. In the case of viruses, this blocks the invader from entering a cell and causing an infection.

Skipping the vaccine

Vaccines trigger the same response. But since there is no vaccine for COVID-19, “we’re skipping the vaccination step and we’re directly making these antibodies,” said Joe Jardine at IAVI, a nonprofit scientific research organization.

Most of the antibodies come from people who have recovered from COVID-19 infection. Their blood is full of them, which is why doctors are testing whether giving COVID-19 patients convalescent plasma — the liquid part of blood from recovered people — could treat or prevent infection.

 

But collecting enough plasma to treat large numbers of patients is labor intensive, and plasma must be screened for other diseases the donor may be carrying.

The blood of recovered patients is also full of the cells that produce antibodies. In recent years, technology has emerged that can sort through millions of these cells to find the ones that produce the most potent antibodies against an individual germ. These cells then form the basis of a manufacturing process to grow large quantities of antibodies.

Patients would receive the treatment by injection or intravenous drip.

So far, two companies have monoclonal antibody therapies undergoing clinical trials for hospitalized COVID-19 patients. At least four others are expected to begin testing this summer.

Antibodies also could be used to provide temporary, vaccine-like protection from infection.

“This would function as a stopgap” for health care workers or other vulnerable people while vaccines are being developed, Jardine said, or as prevention for older people who don’t respond as well to vaccines.

Protection would likely last about a month, possibly six if the antibodies were modified.

Not just one virus

Walker’s group has isolated several antibodies that block not only the coronavirus behind COVID-19 but also the one that caused the 2003 SARS outbreak and a related coronavirus found in bats.

The COVID-19 virus is the third coronavirus in less than two decades to emerge from the wild and kill humans. Many more are circulating in bats and other animals. As people increasingly move into wildlife habitats, humans are coming into contact with a growing number of viruses.

“It’s almost certain we’re going to see something like this happen in the future,” Walker said. “What you want is not just an antibody that works against the current virus, but also against future, related viruses.”

She hopes to start clinical trials later this year.

WHO Aiming for 2 Billion Doses of COVID Vaccine by End of 2021

The World Health Organization’s (WHO) chief scientist said Thursday the agency hopes there will be about two billion doses of a vaccine against COVID-19 by the end of next year that would be reserved for “priority populations.”

Speaking at a virtual news conference in Geneva, Dr. Soumya Swaminathan told reporters, “It’s a big ‘if’ because we don’t have any vaccine that’s proven.”

She said she is encouraged by the number of possible vaccines currently being tested and hoped at least one or two would prove ready for use by next year.

Swaminathan said that the WHO recommends immunizing people at risk first, including the elderly and those with underlying conditions like diabetes or respiratory disease, as well as key workers. But she said countries must come to a consensus on which populations would be prioritized.

Numerous developed countries including Britain, France, the Netherlands, Germany and the U.S. have already struck deals with pharmaceutical firms to secure vaccine supplies for their citizens.

On Wednesday, EU Commission President Ursual von der Leyen announced a donor conference scheduled for June 28 designed to raise funds to ensure all people can get access to any vaccines or COVID-19 treatments that might become available.

She said there is no place for “Me first” when it comes to fighting a global pandemic.

The WHO and partners have called for drug makers to suspend their patent rights on any effective COVID-19 vaccine and for billions of dollars to buy vaccines for developing countries.

COVID School Lockdowns Leading to Increase in Violence Against Children, UN Says

 

WASHINGTON – More and more children are victims of hate, bullying, and violence online because of the coronavirus pandemic, a new United Nations report said.

According to the experts, without access to the support networks such as educators, friends and extended families they usually find at school, some children have been stranded in abusive homes with no place to turn for help as schools are locked down to control the spread of the coronavirus.

Overall, about a billion children suffer from physical, sexual, or psychological abuse each year, especially in places where the governments have failed to set up support programs. COVID-19 isolation is making the problem worse.

“There is never any excuse for violence against children,” said World Health Organization Director General Dr. Tedros Adhanom Ghebreyesus.

In Iran, officials said, regular Friday prayers will resume in Tehran next week despite a jump in the number of coronavirus cases over the past few weeks.

Iran had reported about 198,000 confirmed cases and more than 9,200 deaths as of Thursday, according to trackers at Johns Hopkins University’s coronavirus research center.

Although it still trails the United States, Brazil is quickly becoming the world’s new coronavirus hot spot.

 

Brazil’s health ministry on Thursday reported more than 978,000 cases and 50,000 deaths. According to the Johns Hopkins trackers, the U.S. has just under 2.2 million confirmed cases as of Thursday and more than 118,000 deaths.

Experts at the University of Washington’s School of Medicine told CBS News that according to their model, Brazil’s death toll is poised to surpass that of the United States as early as next month.

Brazil’s President Jair Bolsonaro has dismissed COVID-19 as being nothing more than a “little flu” and said anyone worried about the virus is being neurotic. He has encouraged Brazilian businesses to reopen and states to lift their lockdowns.

The number of confirmed Canadian coronavirus cases climbed to just under 102,000 on Thursday with more than 8,300 deaths, according to Johns Hopkins. Canadian officials said the outbreak appears to be slowing.

While the 13 Canadian provinces and territories are starting to reopen their economies, major restrictions are still in place in the country’s two biggest cities, Montreal and Toronto.

Californians are now required to wear face masks in most indoor and outdoor settings, Gov. Gavin Newsom ordered Thursday.

“Science shows that face coverings and masks work,” Newsom said. “They are critical to keeping those who are around you safe, keeping businesses open and restarting our economy.” The governor said since restrictions started to be lifted last month on restaurants and some stores, not enough people have been taking the proper precautions by covering their faces.

President Donald Trump said in an interview with The Wall Street Journal published Thursday that many people wear masks just to “signal disapproval of him.”

Trump has declined to wear a face mask in public because he said he doesn’t want “to give the press the pleasure of seeing it.” He made that comment last month during a tour of a Ford plant in Michigan, where he said he wore a mask “in the back area.” He has said that he has worn masks away from the cameras at various times.

PRA Health Sciences ready to navigate RACE Act requirements for pediatric oncology

U.S. law requiring clinical research for pediatric oncology treatments goes into effect mid-August 2020

RALEIGH, N.C., June 18, 2020 (GLOBE NEWSWIRE) — Between 2007-2017, 78 novel adult cancer agents were approved by the U.S. Food and Drug Administration,1 but none of those drugs were researched to determine effectiveness for pediatric cancers.

That is about to change on August 18 when the RACE Act – The Research to Accelerate Cures and Equity for Children Act – goes into effect. Passed in 2017, the RACE Act amends the existing PREA – Pediatric Research Equity Act – and eliminates the exemption from PREA requirements for cancer drugs that have orphan status. The new law requires pediatric evaluation of new drugs and biologics intended for the treatment of adult cancers and directed at a molecular target substantially relevant to the growth or progression of pediatric cancer.

Clinical research companies and their pediatric research specialists, including PRA Health Sciences, will be called on to support pharmaceutical and biotech companies as they sponsor the required research for pediatric oncology indications.

  • Cancer drugs approved by the FDA take a median of 6.5 years to go from first clinical trial in adults to the first trial in children4
  • While considered a rare condition, 46 children are diagnosed with cancer each day5 and those patients, their families, and their medical teams need more treatments suitable for pediatric indications

The virtual event, which runs from June 22-25, will incorporate all of the features of a major scientific conference, with keynote speakers, access to CrownBio’s AACR 2020 posters, as well as a tradeshow booth.
